This hypothetical animated special combines elements from the following comic book arcs: Ultimate Knights, Back In Black, and last, but not least, Spider-Men and Spider-Men II.

Plot:
Following the events of 'Spider-Verse', Peter Parker returns to his home universe, relaxes, showers off, and decides to do a web search for Miles Morales. While we don't see the results, Peter can only bring himself to say "Oh my God".

At this point, it's established that Miles Morales and his parents are now in the main NUMAU universe after his home universe got destroyed
[during the events of the Spider-Verse story in our Spectacular Spider-Man season 4 outline].

At the same time, this universe has its own Miles Morales, a former enforcer and assassin who helped Wilson Fisk rise in the underworld as the Kingpin. Miles wanted to quit "the life" and Fisk helped him get out of the underworld, allowing him to settle down in the suburbs.

However, Miles lost his wife Barbara Sanchez and his peaceful suburban life during the events of Spider-Island. After finding out about the multiverse (thanks to research conducted by Dr. Jonathan Ohn, aka the Spot), Kingpin contacts Miles with a proposal: he'll help Miles find an alternate version of his wife, if he kills Spider-Man in return. Miles agrees, taking up the identity he had shelved once again: the Regent.

Peter becomes targeted by Morales
[who throughout I'll be referring to as Miles-B], and naturally, escapes the assassination attempt due to his Spider-sense; unfortunately, his dear Aunt May takes the bullet. Miles-B regrets this, since he doesn't like endangering innocent bystanders.

In response to this, and after a little investigating of his own, Daredevil forms a team of street-level vigilantes called the Knights of New York to bring down the Kingpin once and for all, comprised of Luke Cage, Iron Fist and Jessica Jones, the Punisher (who they manage to convince to 'play it safe' for now), Elektra, Moon Knight and Shang-Chi. Peter and Miles [Miles-A] join Daredevil's team. Miles-A takes up the codename Shadow Spider.

Kingpin, through one of his street-level informants, learns of this development and sends an agent named Ronin to be a mole within the team. The newly-organized group then goes after a number of Kingpin's underbosses, each time shutting down their operation and getting more information that'll take them to the top. At one point, Ronin tries to kill Elektra (in a similar way to Bullseye in Daredevil #181), but fortunately survives thanks to Daredevil's intervention. Ronin is then unmasked by the team, and learn their story.

Her true identity is revealed to be Echo, real name Maya Lopez, a deaf vigilante and martial artist who was using gender-concealing clothes and a voice changer device to disguise herself as Ronin. Maya was tricked by the Kingpin into believing that Daredevil killed her father. Daredevil informs her it was actually the Kingpin who was responsible of her father's death, which helps to redeem Echo and causes her to defect to their side.

Near the end of the battle, the team receives some unexpected help from Miles' own mother Rio Morales, a cop who used to be a SHIELD agent who once went undercover in the organization of the Kingpin (of their former universe). Meanwhile, her husband Jefferson Morales is the doctor who attends to Aunt May while she recovers from her gunshot wounds.

The Kingpin is arrested by Rio after his defeat at the hands of the Knights of New York and both Spider-Men.

Miles-B, after coercing the Spot into working his multiversal apparatus, succeeds in finding an alternate version of his wife Barbara. Miles-B enters the portal leading to that world, and meets her in the very same wine bar she worked at where they first met. They immediately get along swimmingly... until their encounter was interrupted by a fight between Spider-Man and the Trapster...from the
Ultimate Spider-Man animated series.

Meanwhile, back in the main universe, Aunt May recovers just in time to watch Peter's high school graduation.

https://www.imdb.com/name/nm0000984/?ref_=nv_sr_1?ref_=nv_sr_1

Miles worked for Fisk following their time in prison, aiding him during Wilson's usurpation of Don Rigoletto's place as crime boss in New York. Morales took up the moniker of 'the Regent', while Fisk became the Kingpin. Later on, after Miles fell in love with a woman named Barbara Sanchez, Fisk arranged to have all traces of Miles' existence erased from searchable records in order help Miles leave his criminal life behind him. Years later, after Barbara died, Fisk informs a grief-stricken Miles that he has knowledge of a parallel universe in which Barbara might still be alive.
*Since The Spectacular Spider-Man played loose in a few cases of villains--chiefly turning Montana and Fancy Dan of the Enforcers into the Shocker and Ricochet, respectively--I figured slightly merging a couple of other characters couldn't hurt for narrative purposes.*

-The Spot/Dr. Jonathan Ohnn - Danny Jacobs

-Ronin - Tohoru Masamune
https://www.imdb.com/name/nm0556070/?ref_=nv_sr_1?ref_=nv_sr_1

The disguise Echo wears when she infiltrates the Knights. The voice changer in her mask gives those around her the impression of a seasoned male warrior.
